New GPS user and want to now the difference between USA 100K topo and a regional 24K topo. Is the level of detail different?

You will get four times as many contour lines with the 24K mapping. Gives you better elevation resolution.

Why would the 24K have more contourlines than the 100K?

It is a scale. Scale is the relationship between distance on the map and distance on the ground. A map scale usually is given as a ratio such as 1:24K or 1:100K. 1 scale on the map represents 24000 on the ground. In short there will be more lines in 1:24k as per 1:100K. lets say 1 inch for every 24000 inches or in the other 1 inch for every 100000 inches.
